MJFF PPMI iPSC LINES

PPMI is from PPMI participants across different study arms, were used to generate iPSC lines. Lines are available by request at https://www.ppmi-info.org/access-data-specimens/request-cell-lines.

  • MJFF working to generate magnetic controls for these harboring PD mutations.
  • Lines should be available in mid-late 2017.
  • Anyone modifying PPMI iPSC line must deposit the modified line back to the repository this will permit offerings for others moving forward.
  • CSF call test by NF1 CER ALG93 and 2 BMG GFP-NF1 - SK-N-AS

(Autopsy) With autopsy we make a cut through the cerebral peduncles to dissect brainstem and cerebellum from the cerebrum. Then we cut the brainstem in halves. The right half is put into a formaldehyde to later prepare paraffin embedded samples (input superior colliculus, inferior colliculus…). From the left half of the brainstem the substantia nigra and the locus coeruleus are collected in frozen samples. So for most cases we do have 5 frozen and 1 paraffin embedded sample.

Paraffin embedded samples: Upon autopsy, tissue is fixed for about 30 days in 4% formaldehyde. Then the tissue sample is embedded in paraffin. Sample size approximates 1 gram. on request (& depending on committees decision), they may provide the block or they generate FFPE section (8 um, as below).
